Response trajectories

Patients whose tumors responded on imaging (RANO partial or complete response). For each patient, ABP describes the response from that patient's own record: when it began, how the compartments changed, how long it lasted, and whether the scans before it gave any sign. A different patient with a closely matched scan who progressed instead is shown for contrast. Treatment schedules are not recorded in this dataset, so the module describes responses; it does not explain them.

Method and provenance

What the ratings do not include. RANO requires a stable or decreasing corticosteroid dose for a response to count. RANO 2.0 requires confirmation of objective response (after at least four weeks) and requires or recommends confirmation of progression in specified settings, particularly within 12 weeks of radiotherapy because of pseudoprogression. LUMIERE records neither steroid dose nor confirmation status. The ratings shown are the source dataset’s expert RANO labels and have not been re-adjudicated under RANO 2.0; read them as imaging assessments only.
Data. LUMIERE longitudinal glioblastoma dataset (Suter et al., Scientific Data 9:768, 2022): expert RANO ratings and DeepBraTumIA compartment volumes. Response means a RANO rating of partial (PR) or complete (CR) response; it is an imaging assessment, not clinical remission, and pseudo-response or treatment effects are possible.
Response signature. Response period: consecutive PR/CR ratings, ending at the next non-response rating. Changes are measured from the last scan before the response to the lowest enhancing volume within it (enhancing) or to its last scan (edema, necrotic core). Expert ratings use two-dimensional measurements and clinical judgement, so they can disagree with automated volumes; where they do, both are shown.
Before the response. The enhancing growth rate over the interval leading into the last scan before the response, compared with the change at response onset. A decline already under way is reported as a sign; a stable or growing tumor is reported as no sign.
Matched contrast. A scan from a different patient whose next rating was progressive disease, matched on total segmented lesion volume (within 20%), timing, and radiomic imaging state; similarity is the pair's percentile among all cross-patient scan pairs (lower is closer).
3-D view. Where the patient's anatomy file is available, the view shows that patient's reconstructed tumor at the most recent scan, scaled between scans to the interpolated volumes; otherwise equivalent-volume geometry is shown and labelled. Cells are illustrative.
Not recorded. Chemotherapy, radiotherapy and other treatments; operations are inferred from post-operative ratings.
